Yes, the Undo button removes the most recent region. Use Clear to remove every region and start fresh.

4. What's the difference between the three modes?

Pixelate and blur both obscure detail while keeping a rough sense of shape and color; black bar fully blocks the area with a solid rectangle, the most guaranteed way to hide something completely.
